Lipase Immobilized on MCFs as Biocatalysts for Kinetic and Dynamic Kinetic Resolution of sec-Alcohols

نویسندگان

چکیده

Dynamic kinetic resolution (DKR) is one of the most attractive methods for enantioselective synthesis. In reported studies, lipase B from Candida antarctica (CALB) immobilized on siliceous mesoporous cellular foams (MCF) functionalized with different hydrophobic groups, and two ruthenium complexes substituted cyclopentadienyl ligands were investigated as catalysts chemoenzymatic DKR (rac)-1-phenylethanol, using Novozym 435 a benchmark biocatalyst. Studies (rac)-1-phenylethanol transesterification reaction showed that CALB supported MCFs grafted methyl groups promising biocatalyst isopropenyl acetate preferable acylation agent. Both Ru-complexes activated by K3PO4 or t-BuOK, proved to be effective racemization reaction. The final experiments all combinations singled out, gave 96% conversion, (R)-1-phenylethyl enantiomeric excess 98% in 8 h activator.

Double Immobilized Lipase for the Kinetic Resolution of Secondary Alcohols

Sol-gel immobilization of enzymes, which can improve considerably their properties, is now one of the most used techniques. By deposition of the entrapped lipase on a solid support, a new and improved biocatalyst was obtained, which can be used with excellent results in acylation reactions. In this paper, lipase B from Candida antarctica was double immobilized on different adsorbents. These bio...

Lipase-catalyzed kinetic resolution of aryltrimethylsilyl chiral alcohols.

Lipase-catalyzed kinetic resolution of aryltrimethylsilyl chiral alcohols through a transesterification reaction was studied. The optimal conditions found for the kinetic resolution of m- and p-aryltrimethylsilyl chiral alcohols, led to excellent results, high conversions (c = 50%), high enantiomeric ratios (E > 200) and enantiomeric excesses for the remaining (S)-alcohol and (R)-acetylated pro...

Migratory dynamic kinetic resolution of carbocyclic allylic alcohols.

A novel migratory dynamic kinetic resolution based on the interplay between an enzyme acylation catalyst and a heterogeneous Brønsted acid as an isomerization/racemization catalyst gives rise to carbocyclic allylic esters with excellent stereoselectivity from readily available tertiary carbinols. An easy-to-use teabag setup combining resin-bound catalysts, a biphasic isooctane-water solvent sys...

Rapid, room-temperature acylative kinetic resolution of sec-alcohols using atropisomeric 4-aminopyridine/ triphenylphosphine catalysis

Two new atropisomeric 4-aminopyridine-based nucleophilic catalysts containing terphenyl ‘blocking groups’ have been prepared and evaluated for kinetic resolution (KR) of aryl alkyl sec-alcohols. One of these biaryls is shown to be the most selective atropisomeric catalyst yet prepared for several sec-alcohols but its low reactivity makes it non-optimal for use at room temperature (rt). Optimisa...
